| I. Background | |
| cAMP 反應(yīng)元件結(jié)合蛋白 (CREB) 位于細(xì)胞核內(nèi)，是一種轉(zhuǎn)錄因子，在 Ser133 位點(diǎn)被不同的受體活化蛋白激酶（如蛋白激酶 A (PKA)、鈣調(diào)蛋白依賴性蛋白激酶 (CaMK)、絲裂原活化蛋白激酶 (MAPK) 和其他激酶磷酸化后，與靶基因啟動(dòng)子的 cAMP 反應(yīng)元件 (CRE)結(jié)合。CRE 是 CREB 的主要結(jié)合位點(diǎn)，負(fù)責(zé)激活 CREB。CRE 是許多細(xì)胞外和細(xì)胞內(nèi)信號(hào)通路的焦點(diǎn)，包括 cAMP、鈣、GPCR（G 蛋白偶聯(lián)受體）和神經(jīng)營(yíng)養(yǎng)因子。cAMP/PKA 信號(hào)通路對(duì)許多生命過程和生物體至關(guān)重要。在 cAMP/PKA 信號(hào)通路中，CREB 通過 PKA 的磷酸化被激活，并與 CRE 結(jié)合，一般基序?yàn)?5'-TGACGTCA-3'。由于 CRE 是 cAMP/PKA信號(hào)通路的調(diào)節(jié)劑，因此可以研究各種抑制劑的作用。 | |
| II. Description | |
CRE-Luc HEK293 報(bào)告基因細(xì)胞是由 CRE 調(diào)控并表達(dá) Luc 熒光素酶報(bào)告基因的 HEK293 細(xì)胞。Forskolin 是天然存在的直接腺苷酸環(huán)化酶（AC）/cAMP/CREB 的 激活劑，原理見下圖所示。 Figure 1：CRE-Luc HEK293 原理示意圖 |
